연제 : Structural Basis of Cellular Nucleoside Transport

Abstract 

Nucleoside  transporters  (NTs)  are  integral  membrane  proteins  that  are  designed  to  transport  nucleosides  into  the  cells  of  the  human  body.  Nucleoside  transport  across  cell  membranes  is physiologically important as it provides the major source of nucleosides for DNA/RNA synthesis in many cell types, and it is responsible for the termination of adenosine signaling. Furthermore, many nucleoside-­derived  anticancer  and  antiviral  drugs  are  transported  into  cells  by  NTs.  Despite  their  importance in physiology and pharmacology, the molecular mechanism of NTs was largely unknown due to the lack of atomic structures of NTs and structure-­based mechanistic studies. My laboratory has  contributed  significantly  to  our  understanding  of  the  fundamental  mechanism  of  selective  nucleoside transport by concentrative nucleoside transporters (CNTs), one of the two major NTs in human, through structural and functional studies. We anticipate our findings will provide a complete description of nucleoside and nucleoside drug transport across the cell membrane by CNT, which eventually will facilitate the development of new therapeutic reagents.
