Abstract

Intracellular pathogens share a strategy to survive and replicate within a macrophage phagosome by producing virulence proteins. MgtC is one of such proteins that are conserved in intracellular pathogens including Mycobacterium tuberculosis, Salmonella enterica, Yersinia pestis, Brucella suis, and Burkholderia cenocepacia. Being acquired by horizontal gene transfer, genomic location of the mgtC gene differs in each pathogens, however, the mgtC mutants from above intracellular pathogens have a common phenotype: a severe defect in replicating within a macrophage phagosome, suggesting that the MgtC proteins are required for those intracellular pathogens to survive within an acidified phagosomal space. To understand how the MgtC protein helps each pathogen to achieve successful replication inside macrophages, we carried out biochemical and genetic approaches including an in vivo crosslinking experiment followed by immunoprecipitation and a bacterial two-hybrid analysis to search for binding targets. Several proteins are currently identified and investigated to confirm interaction with the MgtC protein, suggesting that MgtC alters pathogen’s bioenergetics and ion transport to promote pathogenicity.
