연제 : Strategy for the production of genetically engineered mouse model

Abstract

A genetically modified mouse model is a mouse that has had its genome altered through the use of genetic engineering techniques. Genetically modified mice are commonly used for research of human diseases and drug development, and are also used for research of function of genes.
Multiple genetic modification techniques have been developed to modify mouse genomes. There are three representative technical approaches to produce genetically modified mice. The first approach is pronuclear injection into a single cell of the mouse embryo, where it will randomly integrate into the mouse genome. The second method is modifying embryonic stem cells with a DNA constructs containing DNA sequences homologous to the target gene. The last method is the genome editing system using engineered nucleases such as ZFN, TALEN, CRISPR/Cas9.
