연제 : Systemic autophagy insufficiency compromises adaptation to metabolic stress and induces progression from obesity to diabetes

Abstract
Despite growing interest in the relationship between autophagy and body metabolism, the role of global autophagy in body metabolism has not been widely studied. We generated mice with global haploinsufficiency of an essential autophagy gene (Atg7+/-mice). While Atg7+/-mice did not show metabolic abnormalities, they developed diabetes when bred to ob/obmice. Atg7+/--ob/obmice showed aggravated insulin resistance with increased lipid content and inflammatory changes, suggesting that autophagy haploinsufficiency impairs adaptive capability to metabolic stress. Intracellular lipid content and insulin resistance after lipid loading were increased in autophagy-insufficiency. Inflammasome was activated in adipose tissue of Atg7+/--ob/obmice. Imatinib administration improved glucose profile and insulin sensitivity in Atg7+/--ob/obmice, which was accompanied by enhanced autophagic flux. These results suggest that systemic autophagy insufficiency could be a factor in the progression from obesity to diabetes. Autophagy modulators may have therapeutic potential against diabetes associated with obesity and inflammation.
