연제 : How to deal with soap crisis: Atomic-level study of membrane protein

Abstract


  Study of the eukaryotic integral membrane proteins is one of the most challenging quest of modern biochemistry and biophysics because of its hydrophobic surface surrounded by membranes, contrast to natively soluble proteins, as well as other problems caused by several posttranslational modifications. So, 2220(less than 700 unique) membrane protein structures have been enrolled in the Protein Data Bank(PDB) and this is just 2% of known atomic-level structures.
  Although more than 50% of drug targets, 40% of all FDA-approved drugs and 20~30% of all proteins in both prokaryotic and eukaryotic organisms are membrane proteins, structural esearch of them is hampered by many hassles and composition of their structures in PDB heavily reflects this kind of difficulties. Nevertheless, more than half of membrane protein structures has been determined during the past six years and development of biophysical and -chemical techniques,for example new detector of EM, eukaryotic expression system and FSEC, keep widening the bottleneck of this field. Thereby this presentation briefly introduces of recent progress of structural study of membrane protein (especially eukaryotic secondary active transporters) and discuss the future directions of this field.
