Abstract

The basal ganglia regulate a broad-range of behaviors, including motor control and cognitive function, and is heavily affected in Parkinson's disease (PD). However, circuit-specific roles of different basal ganglia nuclei and their functional organization has not been fully elucidated. Here, we investigate functional roles of the distinct globus pallidus, external (GPe)-parvalbumin-positive (PV) neurons in motor- and cognitive functions and their contributions to different PD-related behaviors. We demonstrated that substansia nigra pars reticulata (SNr)-projecting GPe-PV neurons and parafascicular thalamus (PF)-projecting GPe-PV neurons are preferentially associated with locomotion and reversal learning, respectively. Finally, we observed that selective manipulation of the SNr-projecting GPe-PV neurons only alleviate motor deficit, whereas manipulation of the PF-projecting GPe-PV neurons only rescued the impaired reversal learning in dopamine-depleted animal. Our findings establish behavioral significance of two distinct GPe-PV neuronal populations and, thereby provide new frameworks for understanding circuit basis of different behavioral deficits in Parkinsonian state.
