Abstract
Safeguarding genomic integrity from genotoxic stresses is critical for biology of a cell. DNA repair or DNA damage responses are under sophisticated controls that must be accurately and rapidly executed, when the genome integrity is challenged. Here I describe two cellular conditions when the genome is vulnerable for obtaining mutations, namely during DNA replication and DNA double strand breaks. In the first part, I describe a new regulatory mechanism that limits DNA replication stress. In the second part, I describe an unexpected role of Polycomb epigenetic regulator in maintaining genomic integrity upon DNA double strand break occurrence.
