Abstract
Many eukaryotic proteins only fold if assisted by the ring-shaped multisubunit chaperonin TRiC/CCT. To understand how TRiC shapes their folding process, we reconstituted the chaperone-assisted folding pathway. ATP hydrolysis induces TRiC chamber closure and tubulin folding. CryoEM structures at near-atomic resolution reveal subunit-specific interactions with the TRiC chamber wall guide through a set of progressively
folded intermediates to reach the native state. A positively charged TRiC wall patch binds both the negatively charged N-terminal tubulin domain. The disordered CCT C-termini within the chamber promote subsequent folding of the core and middle tubulin domains and formation of the GTP-binding pocket. Since all TRiC/CCT-tubulin contact motifs are conserved, it appears CCT subunits coevolved with obligate substrates to acquire extrinsic information to direct folding of novel proteins with unique topological challenges.
